mutant leucyl synthetase + terminator
This part contains a mutated leucyl-tRNA synthetase (leuS) gene, and a terminator (BBa_J61048). The synthetase gene has been reengineered to contain the necessary mutations for the synthetase to use CBZ-leucine and not normal leucine when charging leucyl-tRNA. The catalytic site of the enzyme has been modified to be complementary to CBZ-leucine rather than leucine, and the editing site has been modified to be non complementary to CBZ-leucine while being complementary to normal leucine. This mutated leuS part is necessary for charging leucyl-tRNA synthetase with CBZ-leucine, thereby generating a dependency on CBZ-leucine for the purposes of containing the genetically engineered organism. The strength of expression of this part can be varied in the organism by adding a promoter and RBS of a desired strength. This allows for easy manipulation of this part by future teams to best serve their purposes.
